Involvement of monocyte-derived dendritic cells in the down-regulation of the allogeneic mixed lymphocyte reaction.
In a mixed lymphocyte culture (MLC), activated lymphocytes stimulate mitogen-unresponsive monocytes and subsequently generate dendritic cells (DC) which function to stimulate antigen-specific lymphocytes in primary and secondary responses. The purpose of the present study was to determine the role of monocyte-derived DC in regulating the MLC. Monocytes were enriched with the monocyte-specific monoclonal antibody, IM7, and purified allogenic monocytes and T cells were mixed in an MLC to define the in vivo differentiation of monocytes to DC. In addition to monocyte-derived DC, MLC induced a second population of T-cell stimulatory DC which expressed Ia antigens and were positive for CD1 monoclonal antibodies. The two types of stimulatory DC became apparent 2-4 days after MLC and were detectable for 6-10 days. Treatment of allogeneic monocytes with interferon-gamma (IFN-gamma) increased the level of DC generation by 20-fold, whereas IFN-alpha had no detectable effect. In addition, IFN-gamma inhibited the generation of Lyt2+ monocytes, and anti-IFN-gamma treatment of the monocytes enhanced the allogeneic MLC. The importance of monocyte-derived DC in the down-regulation of the MLC was demonstrated by the inhibition of the generation of stimulator DC by anti-Ia monoclonal antibodies. The ability of monocyte-derived DC to down-regulate the MLC could be mimicked by exposing unseparated lymphocytes to exogenous IFN-gamma in the absence of monocytes. Thus, monocyte-derived DC are probably involved in the regulation of allogeneic T cell responses.-US/en; ; accessed March 20, 2017).

Get issue with Formatter.Parse

I’m trying to construct a new DateTime value from a string, which is basically this: 2011-04-11T00:00:00.000Z.
Here is a code fragment I’m trying to get to work:
String xx = “2011-04-11T00:00:00.000Z”;
DateTime yy = DateTime.ParseExact(xx, “yyyy-MM-dd’T’HH:mm:ss.fff’Z'”,

yy.ToString() always outputs “1/4/2011 12:00:00 AM”. I’ve been trying to google a solution, but haven’t been successful. Any help would be greatly appreciated.

The format string should include the letters uppercase Y (for yyyy) and Z (for Z). This is how you do it in c#.
String xx = “2011-04-11T00:00:00.000Z”;
DateTime yy = DateTime.ParseExact(xx, “yyyy-MM-dd’T’HH:mm:ss.fff’Z'”,

Class constructor not called on instantiation of inherited class

I have inherited the class ConDatabase.
The c’tor of class ConDatabase is not called when instantiating a ConDatabase object, whilst class ConDatabase does contain a c’tor.
The reason for this is because class ConDatabase has a static instance of another class (ConForm).
class ConDatabase :
public ConDatabase()
conForm = new ConForm();

class ConForm :
public ConForm()
// do something
