Two-dimensional LC/MS/MS analysis of human plasma to identify and validate peptides.
The analysis of endogenous peptides is an important research area, due to the possible involvement of peptides as biomarkers in various diseases and for their potential function as drug targets. In this study we have developed a protocol for the identification and quantification of endogenous peptides in human plasma using LC/MS/MS. This strategy was applied to the plasma of two clinically distinct populations: healthy volunteers and patients diagnosed with type 2 diabetes. A list of 4092 human peptides from 453 proteins was identified in these samples. Of these, 114 endogenous peptides were differentially expressed between the two groups, which were further validated using stable isotope labeled analogues, and the resulting data were further confirmed by a second analytical method. Using this strategy, we were able to identify peptides that were not previously reported, even in the UniProt database.The present invention relates to methods and compositions for the delivery of therapeutic compositions into the body, and more particularly relates to polymeric compositions for use in a variety of drug delivery and sensing applications.
