, a larger FOV is preferable to provide a more detailed knowledge of the dynamics of zebrafish heart contraction and allow one to build reliable models of other physiological data.

Conclusion {#s4d}

Our study demonstrates the use of a non-invasive method for evaluating the functional and morphological properties of the zebrafish heart in living animals. We have shown that by monitoring the cardiac activity of zebrafish using the “Minimally Invasive Multiphoton Microscopy” method, it is possible to non-invasively monitor the physiological properties of the zebrafish heart and uncover the capability to survive long-term in vivo imaging at a sub-millimeter scale. The possibility to use long-term non-invasive multiphoton microscopy methods on zebrafish is a novel application that has not been widely explored in this setting.

This study demonstrates that small-scale imaging of the zebrafish heart can yield both *in vivo* and *in vitro* physiological data. Moreover, the technique allows for the measurements of cardiac parameters to be conducted without the need for sophisticated instruments such as echocardiographs. This novel imaging technique will enable us to easily and non-invasively evaluate heart physiology in a wide array of applications, especially in cardiac regeneration studies.

We thank Prof. Michael L. Webb for useful suggestions and comments; Dr. Priyanka Ponnareddy for her contribution on the emergency of the zebrafish lines.
